We are interested in understand the molecular mechanisms of T cell activation by using combination of molecular and imaging techniques. Especially with regard to the early events when T cells make initial contact with the Antigen Presenting Cells APC. The physical contact of T Cell Receptor TCR with pMHC on the surface of APC is translated into a biochemical event inside the T Cells, where the CD3-zeta chain of the TCR complex become phosphorylated by the Tyrosine Kinase Lck. The exact molecular steps of this action is remain to be addressed. As showing in the diagram below, the process potentially involves membrane disassociation of CD3-zeta from inner membrane, TCR clustering and spatial redistribution of TCR, LCK and CD45 at the tips of the microvilli.
